Sterols 1. Cholesterol and Cholesterol Esters

چکیده
In animal tissues, cholesterol (cholest-5-en-3β-ol) is by far the most abundant member of a family of polycyclic compounds known as sterols (see also our web pages on plant sterols, oxysterols, etc.). It can also be described as a polyisoprenoid or a triterpene because of its biosynthetic origin. It was first recognized as a component of gallstones as long ago as 1770, while the great French lipid chemist Chevreul isolated it as a single pure substance in 1815. However, it was well into the 20 century before the structure was fully defined by the German Chemist Heinrich Wieland, who received the Nobel Prize in Chemistry for his work in 1927, the first of many so honoured for research on cholesterol and its metabolism.

Highly sensitive analysis of sterol profiles in human serum by LC-ESI-MS/MS.
We have developed a highly sensitive and specific method for the analysis of serum sterol profiles. Sterols in 1 mul of dried serum were derivatized into picolinyl esters (3beta-picolinate) and analyzed by liquid chromatography-tandem mass spectrometry (LC-MS/MS) using the electrospray ionization (ESI) mode. In addition to cholesterol, 19 cholesterol precursors, cholestanol, campesterol, sitost...
متن کاملSerum, biliary, and fecal cholesterol and plant sterols in colectomized patients before and during consumption of stanol ester margarine.
BACKGROUND Cholesterol metabolic studies are simplified in colectomized patients because of rapid intestinal passage and reduced bacterial action. OBJECTIVE Our objective was to study the effect on cholesterol and plant sterol metabolism of feeding a margarine containing stanol ester to 11 colectomized patients. DESIGN A margarine containing 2 g stanol was consumed for 7-18 d. Serum, biliar...
متن کاملFluorescent Sterols and Cholesteryl Esters as Probes for Intracellular Cholesterol Transport
Cholesterol transport between cellular organelles comprised vesicular trafficking and nonvesicular exchange; these processes are often studied by quantitative fluorescence microscopy. A major challenge for using this approach is producing analogs of cholesterol with suitable brightness and structural and chemical properties comparable with those of cholesterol. This review surveys currently use...
متن کاملSerum sterols during stanol ester feeding in a mildly hypercholesterolemic population.
We investigated the changes of cholesterol and non-cholesterol sterol metabolism during plant stanol ester margarine feeding in 153 hypercholesterolemic subjects. Rapeseed oil (canola oil) margarine without (n = 51) and with (n = 102) stanol (2 or 3 g/day) ester was used for 1 year. Serum sterols were analyzed with gas-liquid chromatography. The latter showed a small increase in sitostanol peak...
متن کاملCholesterol metabolism in type 1 diabetes.
Little information is available on cholesterol absorption and synthesis in human type 1 diabetes. We studied these variables using serum cholesterol precursor sterol ratios to cholesterol as surrogate markers of cholesterol synthesis and those of cholestanol and plant sterols to reflect cholesterol absorption in seven type 1 diabetic subjects and in five age- and body weight-matched control sub...
متن کاملPlant sterol or stanol esters retard lesion formation in LDL receptor-deficient mice independent of changes in serum plant sterols.
Statins do not always decrease coronary heart disease mortality, which was speculated based on increased serum plant sterols observed during statin treatment. To evaluate plant sterol atherogenicity, we fed low density lipoprotein-receptor deficient (LDLr(+/-)) mice for 35 weeks with Western diets (control) alone or enriched with atorvastatin or atorvastatin plus plant sterols or stanols. Atorv...
